Optimization of Expression and Purification of BPSL2774 Protein from Burkholderia Pseudomallei

Description:
I just need the literature review part done for my ongoing final year project.

The target protein, BPSL2774 is a hypothetical protein of the bacteria Burkholderia pseudomallei, which is the causative agent of the infectious disease melioidosis.

Contents my supervisor probably wants to read about:
Melioidosis is indeed a problem and especially so in Malaysia
B. pseudomallei is formidable; it"s resistant to many antibiotics, for instance
The auto-induction process works
The target protein is interesting (I don"t know, maybe)
Below are some details that I"m not sure if relevant in writing a literature review.

Research Objectives:
To obtain the target protein, BPSL2774 in high purity and solubility
To identify possible enzymatic function of BPSL2774 using an enzymatic assay (UDP-Glo Glycosyltransferase Assay, Promega)
BPSL2774 is a target gene from B. pseudomallei that has previously (in a study by my current supervisor"s previous Masters student, unpublished) been predicted as essential by Transposon-Directed Insertion site Sequencing (TraDIS) technique. It has been cloned into Gateway pDEST15 as GST-tagged BPSL2774.

Methods:

1. Cell Culture - Auto-Induction Process
The induction method to be used is the auto-induction process, which F. William Studier thoroughly described in his 2005 report. His studies showed that the method produces higher yields of target proteins in comparison with conventional IPTG induction method.
2. Cell lysis - Sonication
Because we have the machine, apparently.
3. Protein Purification - GST-tagged Affinity Chromatography
4. Observation of Protein Expression - SDS-PAGE
5. Protein Analysis - Functional Assay
If we achieved objective #1.

Finally, I"m attaching my final year project proposal for reference. Although, mind that my examiners were not impressed with it at all, pointing out that my methods didn"t include any kind of optimization and commenting on my rambling/storytelling language style (which I didn"t realize was leaking out onto everything I write). I did realize the former, but I didn"t really know how I was going to do anything at all to begin with and had just gone with what my supervisor had told me I was going to do.
